This study combined single-cell RNA-seq with whole-cell patch-clamp recording to profile the neurons in mouse preoptic area with characterized temperature-sensitivity Overall design: The temperature-sensitivity of 96 neurons in mouse preoptic area was characterized with whole-cell patch-clamp recording in freshly prepared brain slices, following which, the cell samples were harvested and subjected to single-cell RNA seq with an established protcol, Smart-seq2.
