This project used 454 pyrosequencing to sequence partial 18S PCR products from Australian floodplain soil eukaryotes. The 18S PCR products were created using conserved 18S universal rRNA gene primers to amplify a subregion of the 18S rRNA gene from genomic DNA extracted from Australian floodplain soil samples. 454 adapter A and B and barcode sequences were then added to these individual 18S PCR products using 18S forward and reverse fusion primers in a 2nd round of PCR. The intention is to submit the raw 454 sequences (two .sff files from a 2 gasket PTP run) to the SRA using the SRA Interactive Submission Tool, and to submit a subset of ~6000 validated sequences with annotations to GenBank.
