More...Somatic retroelement (RE) insertions are typical for the brain where they were proposed to contribute to somatic mosaicism of neuronal genomes. Although the sequencing capacity of high-throughput sequencing technologies is rapidly growing, studies of somatic RE insertions are still limited to small samples, from one to few hundred cells, mainly due to limitations in enrichment protocols. The goal of this study is the development a new approach for identification and quantification of somatic Alu retroelement insertions. This approach is based on selective amplification of the regions flanking retroelement insertions, normalization of this amplicons using Kamchatka Crab nuclease and sequencing normalized amplicons by Illumina HiSeq
