More...B. bacteriovorus’ genome encodes for one c-di-GMP riboswitch, a standalone highly conserved GEMM motif (c-di-GMP-I riboswitch) not followed by a CDS. It is the most abundant transcript in the AP, represented in transcroptomics by a number of reads similar to that of the total mRNA. It is silenced already upon entry to the TP and steeply reduced by 3 orders of magnitude in the GP. This c-di-GMP aptamer is coined merRNA (massively expressed riboswitch RNA) and is hypothesized to function as a c-di-GMP sink during the AP, sequestering c-di-GMP till phase transition. Here, we deleted merRNA in an otherwise WT B. bacteriovorus HD100.
