History of Nectria haematococca MPVI isolate 77-13-4: This is an isolate that originated from the PhD dissertation work of Harold Kistler 1983 "Phytoalexin metabolism and implications for phytoalexin tolerance and virulence in two plant pathogenic fungi". Cornell University. It is isolated from a tetrad of a third-generation cross from two field isolates in which one parent, T-2, was isolated from infected pea in upstate NY (Genesee Valley) by Gary Harmon. The other parent, T-219, came from the Penn State collection (isolate S-544) and was isolated from the soil from a potato field in Pennsylvania. The Cross 77 was reported in two publications in 1984 (Kistler, H.C. and H.D. VanEtten. 1984. Journal of General Microbiology 130:2595-2603; Kistler, H. C., VanEtten, H. D. 1984. Journal of General Microbiology 130:2605-13.) but isolate 77-13-4 was not specifically mentioned. This isolate is specifically mentioned and shown to have a supernumerary chromosome in the following publication: Wasmann, C. C. and H. D. VanEtten. 1996. Molecular Plant-Microbe Interactions 9: 793-803.
DNA isolation was done as reported in Temporini ED, VanEtten HD (2002) Curr Genet 41: 107-114.
Taxonomy: Nectria haematococca is commonly referred to by its asexual name Fusarium solani and is a member of a monophyletic clade that includes over 50 phylogenetic species known as the "Fusarium solani species complex". Members of the Fusarium solani species complex that cause infections in both humans and plants are common in the environment. The most extensively studied species of the F. solani complex is "mating population" (MP) VI of N. haematococca (also called Haematonectria haematococca). The term "mating population" defines a group of isolates that are sexually fertile with one another, indicating that they are a biological species.
The assembly is ordered and orientated on the optical map. A significant number of smaller scaffolds (7.7% of the total sequence) is not mapped to chromosomes.
