The Atlantic Cod genome was sequenced from a wild-caught individual from the The North East Arctic cod (NEAC) population, designated NEAC_001. Due to the large population size we expected NEAC_001 to exhibit substantial levels of heterozygosity. Genomic DNA used for sequencing was obtained from blood (using high molecular weight DNA agarose blood plugs). Sequencing was done exclusively using the 454 GS FLX Titanium platform and protocols, with both shotgun and paired-end libraries (insert sizes ranging from 1 to 20 kb). We calculated genome size, based on average sequencing depth (k-mer distributions), to be 830 Mbp. The raw reads were assembled using newbler from 454 Life Sciences, and numerous gaps due to heterozygosity were resolved using custom scripts.
