[ELAC2] The protein encoded by this gene has a C-terminal domain with tRNA 3and#8242; processing endoribonuclease activity, which catalyzes the removal of the 3' trailer from precursor tRNAs. The protein also interacts with activated Smad family member 2 (Smad2) and its nuclear partner forkhead box H1 (also known as FAST-1), and reduced expression can suppress transforming growth factor-beta induced growth arrest. Mutations in this gene result in an increased risk of prostate cancer. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq].

Expression: According to AceView, this gene is expressed at very high level, 4.9 times the average gene in this release. The sequence of this gene is defined by 553 GenBank accessions from 516 cDNA clones, some from brain (seen 46 times), tongue, tumor tissue (29), testis (27), amygdala (17), cerebellum (17), thalamus (17), lymph (13) and 127 other tissues.
Alternative mRNA variants and regulation: The gene contains 38 distinct introns (36 gt-ag, 2 gc-ag). Transcription produces 21 different mRNAs, 18 alternatively spliced variants and 3 unspliced forms. There are 5 probable alternative promotors, 3 non overlapping alternative last exons and 14 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 3 cassette exons, overlapping exons with different boundaries, splicing versus retention of 12 introns. 878 bp of this gene are antisense to spliced gene RICH2, raising the possibility of regulated alternate expression.
2 variants were isolated in vivo, despite the fact that they are predicted targets of nonsense mediated mRNA decay (NMD).
Function: There are 39 articles specifically referring to this gene in PubMed. Functionally, the gene has been tested for association to diseases (OMIM: Prostate cancer; Other sources: Genetic Predisposition to Disease; Prostate cancer, susceptibility to; Prostatic Neoplasms) and proposed to participate in a process (tRNA processing). Proteins are expected to have molecular functions (protein binding, endonuclease activity, hydrolase activity, metal ion binding) and to localize in various compartments (endoplasmic reticulum, membrane, mitochondrion, nucleus). Putative protein interactors have been described (ASCC2, CTBP1, CTBP2, CUX1, ISCA1ANDC9ORF153ANDGOLM1, NDUFA7ANDCD320, PUF60, RING1).
Protein coding potential: 18 spliced mRNAs putatively encode good proteins, altogether 19 different isoforms (10 complete, 3 COOH complete, 6 partial), some containing Metallo-beta-lactamase superfamily domain [Pfam]. The remaining 3 mRNA variants (3 unspliced) appear not to encode good proteins. Finally proteins from this gene may be modulated by acetylation; phosphorylation; ubiquitination, as detailed at PhosphoSite.
