SRX130317: Transcriptome sequencing and annotation of the predatory mite Metaseiulus occidentalis (Acari: Phytoseiidae): a cautionary tale about possible contamination by prey sequences
1 LS454 (454 GS FLX) run: 785,142 spots, 412.1M bases, 927.4Mb downloads

Accession: SRX130317
Experiment design: Total RNA was transcribed to cDNA with an oligo dT primer, with second strand then generated. The cDNA is digested with a restriction enzyme then adaptors ligated onto the double-stranded (ds) DNA. The adaptor ligated cDNA fragments were the amplified by PCR. This product was purified and processed for emPCR and 454 sequencing according to the manufacturer's recommended methods.

Abstract: RNA sequencing was performed from mixed tissues of Metaseiulus occidentalis on the 454 platform, assembled, and submitted as TSAs. Transcripts for genes possibly involved in diapause and foraging also were identified, as well as putative genes associated with pesticide resistance. Thus, novel and important genes were discovered that provide insight into the evolution, biology, and physiology of this important predator of pest mites in agriculture.
