Design: We collected deep subseafloor crustal fluids from borehole observatories located on the Juan de Fuca Ridge flank. Fluids were filtered to collect the microbial size-fraction and preserved at -80 C until transport to the laboratory. DNA was extracted from the samples and the SSU rRNA genes were amplified from the environmental genomic DNA using a barcoded primer (515F/806R) approach to perform sequencing of multiplexed samples (Caporaso et al., 2011; PNAS).
