Bristle MTs are polarized minus end out. Confocal projections of bristles containing Nod-KHC-β-Gal (A), GFP-Nod-KHC:neur-Gal4 (B), or kinesin-β-Gal:neur-Gal4 (C), stained with anti-β-Gal antibodies (green) (A and C), Alexa fluor phalloidin (red) (A to C), or fluorescent Hocest (blue) (C). The MT minus-end marker, Nod-KHC, is localized to the tip in wild-type bristles (A and B), whereas the MT plus-end marker, kinesin, is found at the bristle base (C), where it is localized in the nucleus (C, inset). The fluorescent signal intensity for Nod-KHC-β-Gal (A′), GFP-Nod-KHC:neur-Gal4 (B′), or neur-Gal4; pUAS kinesin-β-Gal (C′), as measured along each of the bristles, respectively, is shown. Scale bar, 5 μm.
