Affinity of single-headed Ncd is significantly lower compared with dimeric motor. The conditions used are as follows. 0.4 μm 1,5-IAEDANS-labeled Ncd or NcN was incubated for 15 min with microtubules at the indicated concentration in 20 mm Hepes/KOH, 100 mm potassium acetate, 5 mm magnesium acetate, 0.1 mm EGTA, 0.1 mm EDTA, 5% sucrose, 10 μm dithiothreitol, pH 7.2 (25 °C, no external nucleotide). The reaction mixture was centrifuged at 80,000 rpm for 30 min, the supernatant was collected, and the concentration of Ncd was determined from the fluorescence. The values are mean ± S.D.
