Changes in gene expression of Neurog1, Brn3a, GATA3, NeuroD1, Ntrk1, Ntrk2, Ntrk3 and GFRα1 were analyzed at 72h and 5D relative to uninduced cells at 0h (i.e. undifferentiated cells). qRT-PCR confirmed Dox initiated Neurog1 expression, which results in large upregulations of Ntrk2 and Ntrk3 at both time points analyzed. Ntrk1 had a smaller upregulation at 72h that was no longer present at 5D. GFRα1 was also upregulated at 72h and 5D. After termination of Neurog1 expression, Brn3a, GATA3, and NeuroD1, which are all transiently upregulated by Neurog1 during normal SGN development, decreased significantly between 72h and 5D. The expression profiles of these genes suggest SGN differentiation may be initiated in ES cells forced to express Neurog1 for 48h.
