Determination of an equilibrium constant for efavirenz for RT–T/P. (A) Burst reactions carried out in the presence of different of 0 (•), 2 nM (▾), 4 nM (Δ), 7.5 nM (○), 15 nM (▪), and 35 nM (□) EFV, respectively. The final concentrations of RT, T/P, TTP, and Mg2+ in the assay were 50 nM, 120 nM, 20 μM, and 10 mM, respectively. (B) The amplitudes of the burst phases (•) were plotted against EFV concentration and fit to a hyperbolic function (solid line) to yield a Kd value of 2.6 ± 1.3 nM. Similar experiments were conducted to determine Kd values for NEV and DEL (data not shown).
