WIPI49 binds to PtdIns3P, PtdIns3,5P2, and PtdIns5P in vitro. (A) Liposomes of defined phosphoinositide content were generated as described in MATERIALS AND METHODS and subsequently incubated with purified GST-WIPI49 or GST-SNX3-PX (∼250 ng) for 10 min at 37°C. Resultant lipid complexes were pelleted by centrifugation, separated from supernatants, and resolved by SDS-PAGE and Western blotted using GST-specific antisera.
