Proposed schematic showing the crosstalk of phosphorylation, sumoylation, and ubiquitylation in ATM-regulated signaling in response to DNA damage. Double-strand breaks activate ATM to induce downstream signaling cascade involving sumoylation and ubiquitylation of proteins participating in DNA damage response, such as checkpoint control and DNA repair. STUbL functions as a signal transducer to coordinate SUMO and Ub signals to catalyze SUMO-targeted ubiquitylation, which may recruit p97–UFD1–NPL4 segregase complex to mediate the proteasomal degradation.
