(B) Alternative PMB plate assay of the PAK ΔphoPQ strain expressing the phoPQ+, phoPQ6, or phoP+ allele inserted in plasmid pJN105D. (C) Alternative PMB plate assay of chromosomal deletion mutants PAK ΔphoQ and PAK ΔpmrAB ΔphoQ as well as PAK ΔphoQ expressing the phoPQ+ alleles inserted in plasmid pJN105D. (D) Alternative PMB plate assay of the PAK ΔphoPQ strain expressing mutant phoPQ alleles from CF clinical isolates in plasmid pJN105D. The phoP+Q21 and phoP+Q24 alleles encode deletions from PhoQ of 2 and 276 amino acids, respectively, while the phoP24Q24 allele also encodes replacement in PhoP of Arg 118 with Cys.
