[WNK2] The protein encoded by this gene is a cytoplasmic serine-threonine kinase that contains cysteine in place of the lysine found at the conserved ATP-binding location in subdomain II of protein kinases. Since this protein does have kinase activity, it is possible that another lysine in the kinase subdomain I can substitute for the missing conserved lysine. [provided by RefSeq].

The sequence of this gene is defined by 201 GenBank accessions from 169 cDNA clones, some from brain (seen 19 times), eye (19), hippocampus (10), colon (9), cerebellum (8), lymph (7), uterus (7) and 78 other tissues. We annotate structural defects or features in 31 cDNA clones.
Alternative mRNA variants and regulation: The gene contains 49 distinct introns (47 gt-ag, 2 gc-ag). Transcription produces 28 different mRNAs, 22 alternatively spliced variants and 6 unspliced forms. There are 12 probable alternative promotors, 5 non overlapping alternative last exons and 7 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 8 cassette exons, overlapping exons with different boundaries, splicing versus retention of 3 introns. 1150 bp of this gene are antisense to spliced gene C9orf129, 752 to lortoby, raising the possibility of regulated alternate expression.
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant jAug10).
Function: There are 13 articles specifically referring to this gene in PubMed. Functionally, the gene has been tested for association to diseases (Hypertension; Meningeal Neoplasms) and proposed to participate in processes (intracellular protein kinase cascade, protein amino acid phosphorylation). Proteins are expected to have molecular functions (protein binding, ATP binding, nucleotide binding, protein serine/threonine kinase activity, transferase activity) and to localize in various compartments (membrane, nucleus). A putative protein interactor has been described (ATXN1).
Protein coding potential: 17 spliced and 2 unspliced mRNAs putatively encode good proteins, altogether 19 different isoforms (6 complete, 5 COOH complete, 8 partial), some containing domains Protein kinase domain, Protein tyrosine kinase [Pfam], some transmembrane domains, a N-myristoylation domain, a peroxisomal domain, a second peroximal domain, a coiled coil stretch [Psort2]. The remaining 8 mRNA variants (5 spliced, 3 unspliced; 5 partial) appear not to encode good proteins.
