  The myocyte enhancer factor-2 (MEF2) binding domain, as found in the calcineurin-binding protein cabin-1, adopts an amphipathic alpha-helical structure, which allows it to bind to a hydrophobic groove on the MEF2S domain, forming a triple-helical interaction. Interaction of this domain with MEF2 causes repression of transcription. Cabin-1 inhibits calcineurin-mediated signal transduction in T-cell receptor-mediated signalling pathways, by binding to the activated form of calcineurin. Cabin-1 acts as a co-repressor of MEF2, the mycocyte enhancer factor-2, which regulates transcription in a calcium-dependent manner and plays vital roles in T-cell development and function.
