Label |
DyLight-649-conjugated goat anti-mouse IgM antibody
Label protocol |
Commercially available DyLight 649-conjugated goat anti-mouse antibody was added to the microarray at 1:2000 in 5% fetal bovine serum in phosphate-buffered saline with 0.1% Tween-20.
Hybridization protocol |
Murine antiserum was probed on the antigen microarray at 1:250 dilution in 5% fetal bovine serum in phosphate-buffered saline with 0.1% Tween-20, and incubated for 1.25 hours at 4 degrees Celsius.
Scan protocol |
Fluorescent array images were collected at 635 nm with a GenePix 4000B fluorescent scanner at 10 micron resolution and image intensity data were extracted and analyzed with GenePix Pro 6.1 analysis software.
Description |
313T Murine antiserum was obtained from blood collected from an individual MLR/lpr mouse exhibiting lupus-like symptoms. A 1:250 dilution of this antiserum was then applied onto the Utz Lab Whole Protein Autoantigen Array (V2.0), labeled with a goat anti-mouse IgM secondary antibody conjugated to DyLight-649 (Jackson ImmunoResearch), and scanned with a GenePix 4000B scanner using GenePix Pro 6.1.

Data processing |
Data were background corrected and flagged values were removed. The remaining replicate values were averaged.
