According to manufacturer's protocols. In brief, 70ul of 1,500 fold diluted Fluorescent dye-conjugated streptavidin ws added to each subarray and incubated in a dark room at room temperature for 1-2 hours.

Hybridization protocol |
The array consisted of three glass slides (array 6, 7, and 8) that were pretreated according to the manufacturer's instructions and incubated with 2-fold diluted serum pools for 2 hours. All sample measurements were performed in duplicate. The array glass slides were washed, incubated with a biotin-conjugated anti-cytokine mix for 2 hours, washed again, and developed for 2 hours with Cy3-conjugated streptavidin.

Fluorescence intensities (Cy3) were recorded with an Axon GenePix 4000B scanner.

Data processing |
Raw data was analyzed with the Raybiotech analysis tool, a data analysis program based on Microsoft Excel technology specifically designed for the analysis of these microarrays. Signals were normalized using internal, positive and negative controls included on the array. Signals were normalized using positive and negative control features on the microarray, the fold changes are relative expression to positive control features, i.e. a value of 1 is same expression than positive control, 2 a 2 fold higher, 0.5 a 2-fold lower expression.
