Sample GSM647225 Query DataSets for GSM647225
Status Public on Mar 05, 2012
Title p53_ChIP_seq_Adr8h
Sample type SRA

Source name embryonic stem cells, treated, p53 ChIP
Organism Mus musculus
Characteristics background/strain: 129X1 x 129S1
cell type: embryonic stem (ES) cells
passages: less than 15
agent: adriamycin
chip antibody: anti-p53
antibody vendor: Santa Cruz
antibody catalog #: sc-1312
antibody lot #: K1907
Treatment protocol Cells were either untreated or treated with 0.5 micromolar adriamycin for 8 hours in growth medium.
Growth protocol Cells were grown in mESC growth medium, Knockout DMEM (KO-DMEM), 15% Fetal Bovine Serum, 100mM MEM non-essential amino acids, 0.1mM 2-mercaptoethanol, 2mM L-glutamine and 1X penicillin-streptomycin (All from Invitrogen) plus 1000 units/ml leukemia inhibitory factor (Lif, Millipore).
Extracted molecule genomic DNA
Extraction protocol DNA was extracted with the phenol/chloroform approach. ChIP assays were performed according to the Agilent mammalian ChIP-chip protocol (version 10, May 2008). Library construction was performed as per Illumina's ChIP-seq library construction protocol.

Library strategy ChIP-Seq
Library source genomic
Library selection ChIP
Instrument model Illumina Genome Analyzer II

Description Treated with adriamycin, chromatin IP against p53.
Data processing Short reads were mapped to build mm9 using Illumina's pipeline. Only reads that passed the quality filtering and are unique are retained.
