Treatment protocol |
Test diet blends were prepared by mixing low- or high-CLA beef at a 36% inclusion rate to equal portions of wheat feed and maize (corn) feed. Final feeds were prepared by mixing 100ml warm water with 100g test diet blend. Dietary food intake was measured daily and the animals received freshly prepared food each day.

Growth protocol |
4 week old male, ob/ob (C57BL/6J) mice were exposed to 12 hrs light/ 12 hrs dark cycles, and maintained at a constant temperature of 22 degrees Celcius.

Extraction protocol |
RNA was later extracted using a Qiagen Rneasy extraction kit, according to the manufacturer extraction protocol.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 2µg total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 7G.
