The key paradigms and approaches in studying TRPC regulation by auxiliary proteins. 1, TRPC channel interaction with auxiliary proteins can be probed using Y2H and verified using immunoprecipitation (IP) and FRET with the native or co-expressed proteins. 2, TRPC channel activity can be monitored by patch clamp current recording or fluorescent Ca2+ measurements. 3, The delivery of TRPC channels to the PM can be dynamically monitored using TIRF and FRAP.
