Soluble fraction of ROS contain a CK2 α-subunit variant. Human recombinant casein kinase II (lane a), bovine ROS (lane b), ROS cytosol (lane c), solubilized ROS membranes (lane d), unbound (lane e) and bound (lane f) fraction from PMc 1D1-Sepharose were subjected to SDS gel electrophoresis and either stained with Coomassie Blue or transferred to Immobilon-P membranes and labeled with an anti-CK2a monoclonal antibody. Lanes contained either 40 mg of ROS or 3 mg of purified channel. CK2 α-subunit is present in the soluble fraction from hypotonically lysed ROS and migrates with a slightly higher apparent molecular weight relative to recombinant CK2 α.
