Calibration of a Nafion-coated carbon fiber electrode in room temperature (22°C) phosphate buffered saline (PBS). To determine the selectivity produced by the Nafion film, the electrode was challenged with 250 μM 5-HIAA before the first addition of 5-HT. The linearity of response to 5-HT over the range of 0.0–3.0 μM is excellent (r2 = 0.9956-solid line) but diminishes as the concentration of 5-HT is increased (r2 = 0.9858-dashed line). The selectivity of this particular sensor for 5-HT against 5-HIAA 3544:1 and the detection limit for 5-HT is 29 nM (signal-noise-ratio = 3).
