The RUNX1 protein is biotinylated and CBFβ-SMMHC protein has a hits-tag on its c-terminus. The streptavidin coated donor bead and nickel-chelate acceptor bead capture both proteins, which allows energy transduction from the donor bead to acceptor bead. Binding of an inhibitor to either RUNX1 or CBFβ-SMMHC protein can disrupt the interaction and thus reduces the luminescence signal.
