[FMNL2] This gene encodes a formin-related protein. Formin-related proteins have been implicated in morphogenesis, cytokinesis, and cell polarity. Alternatively spliced transcript variants encoding different isoforms have been described but their full-length nature has yet to be determined. [provided by RefSeq].

RefSeq annotates one representative transcript (NM included in AceView variant.a), but Homo sapiens cDNA sequences in GenBank, dbEST, Trace and SRA, filtered against clone rearrangements, coaligned on the genome and clustered in a minimal non-redundant way by the manually supervised AceView program, support at least 8 spliced variants.

Expression: According to AceView, this gene is expressed at high level, 2.8 times the average gene in this release. The sequence of this gene is defined by 331 GenBank accessions from 297 cDNA clones, some from brain (seen 32 times), hippocampus (14), placenta (14), hypothalamus (11), lung (10), breast (9), head neck (8) and 101 other tissues. We annotate structural defects or features in 15 cDNA clones.
Alternative mRNA variants and regulation: The gene contains 32 distinct introns (29 gt-ag, 3 gc-ag). Transcription produces 12 different mRNAs, 8 alternatively spliced variants and 4 unspliced forms. There are 2 probable alternative promotors, 3 non overlapping alternative last exons and 3 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 4 cassette exons, overlapping exons with different boundaries, splicing versus retention of one intron. 522 bp of this gene are antisense to spliced gene kluneybo, raising the possibility of regulated alternate expression.
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant aAug10).
Function: There are 7 articles specifically referring to this gene in PubMed. Functionally, the gene has been proposed to participate in processes (actin cytoskeleton organization, cellular component organization). Proteins are expected to have molecular function (actin binding) and to localize in cytoplasm. A putative protein interactor has been described (PRPF40A).
Protein coding potential: 7 spliced mRNAs putatively encode good proteins, altogether 7 different isoforms (1 complete, 4 COOH complete, 2 partial), some containing domains Diaphanous FH3 Domain, Diaphanous GTPase-binding Domain, Formin Homology 2 Domain [Pfam], a N-myristoylation domain, a second peroximal domain, a coiled coil stretch [Psort2]. The remaining 5 mRNA variants (1 spliced, 4 unspliced; 2 partial) appear not to encode good proteins. Finally proteins from this gene may be modulated by acetylation; mono-methylation; phosphorylation; ubiquitination, as detailed at PhosphoSite.
