[ANAPC4] A large protein complex, termed the anaphase-promoting complex (APC), or the cyclosome, promotes metaphase-anaphase transition by ubiquitinating its specific substrates such as mitotic cyclins and anaphase inhibitor, which are subsequently degraded by the 26S proteasome. Biochemical studies have shown that the vertebrate APC contains eight subunits. The composition of the APC is highly conserved in organisms from yeast to humans. The exact function of this gene product is not known. [provided by RefSeq].

RefSeq annotates one representative transcript (NM included in AceView variant.b), but Homo sapiens cDNA sequences in GenBank, dbEST, Trace and SRA, filtered against clone rearrangements, coaligned on the genome and clustered in a minimal non-redundant way by the manually supervised AceView program, support at least 13 spliced variants.

Expression: According to AceView, this gene is expressed at high level, 3.3 times the average gene in this release. The sequence of this gene is defined by 380 GenBank accessions from 345 cDNA clones, some from uterus (seen 18 times), placenta (17), well-differentiated endometrialadenocarcinoma, 7 pooled tumors (12), brain (8), kidney (8), breast (5), heart (5) and 93 other tissues. We annotate structural defects or features in 4 cDNA clones.
Alternative mRNA variants and regulation: The gene contains 32 distinct gt-ag introns. Transcription produces 15 different mRNAs, 13 alternatively spliced variants and 2 unspliced forms. There are 6 probable alternative promotors, 2 non overlapping alternative last exons and 5 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 3 cassette exons, overlapping exons with different boundaries, splicing versus retention of 7 introns.
Note that mRNA .gAug10 was found in vivo, although it is a predicted target of nonsense mediated mRNA decay (NMD).
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant cAug10, dAug10, eAug10, gAug10, kAug10).
Function: There are 22 articles specifically referring to this gene in PubMed. Functionally, the gene has been proposed to participate in pathways (Cell cycle, Oocyte meiosis, Progesterone-mediated oocyte maturation, Ubiquitin mediated proteolysis) and processes (cell cycle, cell division). Proteins are expected to localize in various compartments (cytoplasm, extracellular space). Putative protein interactors have been described (ANAPC1, ANAPC2, ANAPC5, ANAPC10, APC2, ARPC3ANDANAPC7, CDC16, CDC23, CDC27, CDH1, FZR1).
Protein coding potential: 12 spliced mRNAs putatively encode good proteins, altogether 12 different isoforms (7 complete, 2 COOH complete, 3 partial), some containing a vacuolar domain [Psort2]; 2 of the 7 complete proteins appear to be secreted. The remaining 3 mRNA variants (1 spliced, 2 unspliced; 1 partial) appear not to encode good proteins. Finally proteins from this gene may be modulated by acetylation; phosphorylation; sumoylation; ubiquitination, as detailed at PhosphoSite.
